Abstract:Objective To detect the expression of candidate tumor suppressor gene TESTIN and RUNX3 in human colorectal carcinoma,and to explore its relationship to clinical and pathological features of colorectal carcinoma.Methods The expression of TESTIN and RUNX3 was detected by immunohistochemistry in 60 colorectal carcinoma specimens and their corresponding adjacent normal tissues.Results The rates of positive expression and strong positive expression of TESTIN in colorectal cancer tissues were significantly lower than those in adjacent normal tissues(31.7% and 5.0% vs 86.7% and 60.0%,respectively;P<0.01).In addition,the rates of positive expression and strong positive expression of RUNX3 in colorectal cancer tissues were significantly lower than those in adjacent normal tissues(26.7% and 1.7% vs 81.7%and 55.0%,respectively;P<0.01).The expression of TESTIN and RUNX3 was positive correlated with infiltration,poor differentiation,Dukes stage(C and D),T stage(T3 and T4),lymph node metastasis and distant metastasis(r=2.352,1.485,1.867,2.541,0.885 and 3.214,respectively;P<0.05),but not with gender,age and carcinoma location(r=-0.452,-0.245 and -0.331,respectively;P>0.05).Furthermore,there was a positive correlation between TESTIN expression and RUNX3expression(r=6.587,P<0.05).Conclusion Both TESTIN and RUNX3are closely related to biological behaviors of colorectal carcinoma,and may be valuable as markers for assessing the prognosis and metastasis of colorectal carcinoma and as new targets for treating colorectal carcinoma.
