Abstract:Objective To evaluate the application value of immunohistochemistry combined with in situ hybridization for detecting immunoglobulin κ,λ light chain restriction in mantle cell lymphoma(MCL).Methods The expression of CyclinD1 was detected by immunohistochemistry using Envision two step method in 15 cases of nodular mantle cell lymphoma(group 1),15 cases of diffuse mantle cell lymphoma group(group 2) and 10 cases of reactive lymph node hyperplasia(control group).The κ to λ light chain ratio was measured by in situ hybridization to determine the light chain restriction of tumor cells.Results CyclinD1 was positively expressed in nodular mantle cell lymphoma and diffuse mantle cell lymphoma(15/15,100.0%),but was not expressed in the surrounding residues or lymphocytes with reactive hyperplasia.In group 1,κ light chain restriction was found in 7 cases(7/15,46.7%),λ light chain restriction in 3 cases(3/15,20.0%),and no light chain restriction in 5 cases(5/15,33.3%).In group 2,κ light chain restriction was found in 2(2/15,13.3%),λ light chain restriction in 6 cases(6/15,40.0%),and no light chain restriction in 7 cases(7/15,46.7%).No cases showed light chain restriction in control group.Conclusion The light chain restriction and types can be detected by cyclin D1 immunohistochemistry and in situ hybridization in different types of MCL.Therefore,the combined detection is important for early diagnosis of MCL.
