Abstract:Objective To elucidate the role of haematopoietic cell-specific protein1-associated protein X-1(HAX1) on chemotherapy-induced apoptosis in SW480 cells.Methods The pcDNA-HAX1 plasmids were constructed and transiently transfected into in SW480 cells(HAX1 group).Additionally,cells in CTL group were transfected with control empty plasmids.Twenty-four hours after transfection,cells were treated with cis-platinum(10 μg·mL-1) for 36 hours.The apoptosis was detected by Sub-G1 and annexin V-FITC assay.The protein expression of HAX1,caspase-9 and PARP was determined by Western blot.Results After 36 hours of cis-platinum treatment,sub-G1 and annexin V-FITC results showed that the apoptosis rates in CTL group were significantly higher than those in HAX1 group [(33.67±2.50)% vs(16.33±2.10)%,P=0.026 and(46.33±9.30)% vs(24.33±6.10)%,P=0.013,respectively).Western blotting showed that the overexpression of HAX1 obviously inhibited the cleavage of both caspase-9 and PARP.Conclusion The overexpression of HAX1 inhibits SW480 cell apoptosis induced by cis-platinum and the mechanism is involved in the decrease in caspase-9 and PARP cleavage.
