Expression of Kv1.3 in Celiac Macrophages and Influence of Low Potassium on Macrophage Function in Continuous Ambulatory Peritoneal Dialysis-related Periton
Abstract:Objective To explore the expression of Kv1.3 in celiac macrophages and the influence of low potassium on macrophage function in continuous ambulatory peritoneal dialysis(CAPD)-related peritonitis.Methods Thirty patients with CAPD-related peritonitis hospitalized in Jiangxi province people’s hospital between October 2009 and August 2010 were selected as experimental group,and 30 CAPD patients without peritonitis as control group.The expression of Kv1.3 in celiac macrophages was detected by flow cytometry.RAW 264.7 macrophages were cultured in RPMI1640 medium supplemented with different concentrations of K+(5.4,2.0,4.0 and 10.0 mmol·L-1K+).The LDH,neutral red phagocytosis and E.coli colony forming units(CFU) were determined to observe the effects of low K+ on macrophage activity,phagocytic capacity and bactericidal ability.Results The expression of Kv1.3 in effusion macrophages in experimental group was lower than that in control group,but the difference was not statistically significant[mean fluorescence intensity:(8.78±2.30) % vs(9.54 ± 3.61)%,P>0.05].The levels of LDH in 5.4 mmol·L-1K+ treatment group were significantly different from those in 2.0,4.0 or 10.0 mmol·L-1K+ treatment group after culture for 4,8,12 and 24 hours(all P<0.05).Furthermore,LDH levels after culture for 4 hours were obviously different from those after culture for 8,12 or 24 hours in 2.0,4.0 and 10.0 mmol·L-1K+ treatment groups(all P<0.05).Compared with 5.4 mmol·L-1K+,neutral red phagocytosis was significantly decreased by 2.0 mmol·L-1 K+ treatment(0.21±0.02 vs 0.15±0.01,P<0.01),but was not changed by 4 and 10.0 mmol·L-1K+ treatment(0.21±0.02 vs 0.19±0.03 and 0.22±0.05,all P>0.05).Compared with 4.0 mmol·L-1K+,neutral red phagocytosis was significantly decreased by 2.0 mmol·L-1K+ treatment(0.19±0.03 vs 0.15±0.01,P<0.01).Compared with 10.0 mmol·L-1K+ treatment,neutral red phagocytosis was significantly decreased by 2.0 and 4.0 mmol·L-1K+ treatment(0.22±0.05 vs 0.15±0.01 and 0.19±0.03,P<0.01).In addition,the number of bacteria phagocytized and number of bacteria killed in 2.0 mmol·L-1K+ treatment group were significantly lower than those in 5.4 mmol·L-1K+ treatment group(P<0.05),and these numbers in 2.0 or 4.0 mmol·L-1K+ treatment group were obviously lower than those in 10.0 mmol·L-1K+ treatment group(P<0.01).Conclusion The expression of Kv1.3 in celiac macrophages is not obviously changed in CAPD-related peritonitis.But low potassium can significantly reduce phagocytic capacity and bactericidal ability of macrophages.
