Abstract:Objective To investigate the effect of hyperbaric oxygen(HBO)on the expression of matrix metalloproteinase-9(MMP-9)in rat brain tissue in cerebral ischemia-reperfusion injury.Methods Forty SD rats were randomly divided into HBO treatment group(group A,n=20) and high pressure air treatment group(group B,n=20).Each group was further subdivided into reperfusion model group(group A1and group B1,n=10)and sham-operation group(group A2 and group B2,n=10).Cerebral ischemia-reperfusion model was established using suture method.Brain tissue water content and blood-brain barrier permeability were determined by dry/wet weight method and Evans blue staining,respectively.The expression of MMP-9 was measured by ELISA and Western Blot.Results Compared with group B2,brain tissue water content,bloodbrain barrier permeability and MMP-9expression were significantly increased in group B1(P< 0.05).Compared with group A1,these parameters slightly decreased in group A2(P>0.05),but significantly increased in group B1(P<0.05).Conclusion The increase in MMP-9expression is correlated with blood-brain barrier injury and brain edema after cerebral ischemia-reperfusion injury.HBO treatment can inhibit the expression of MMP-9,maintain blood-brain barrier integrity,and relieve brain edema.
