Abstract:Objective To investigate the inhibition effect of cytosine deaminase(CD) gene-modified neural stem cells(NSCs) and the action mechanism,and to provide a theoretical basis for the treatment of glioma.Methods Neurosphere-forming NSCs were isolated from fetal brain.CD gene was introduced into NSCs by liposome-mediated transfection and placed under G418 selection.NSCs/CD and C6 glioma cells were co-cultured and cell survival was assessed with MTT assay.Results CD gene-modified NSCs were successfully constructed.NSCs/CD significantly inhibited the growth of C6 cells.Conclusion NSCs are good carrier cells and NSCs/CD-5-FC suicide gene therapy system has a strong anti-tumor effect.
